Natural killer cell-mimicking nanomaterial for overcoming the multidrug resistance of tumor via cascade catalysis

چکیده

Despite the availability and prosperity of chemotherapeutic agents, multidrug resistance (MDR) remains paramount obstacle in antitumor chemotherapy. Here, a natural killer (NK) cell-biomimetic nanoparticle (DMLN) is developed to overcome MDR by promoting entry doxorubicin (DOX) into tumor cells via cascade catalysis. DMLN synthesized compounding NK cell membrane with hollow MnO2 nanoparticles loaded DOX lactate oxidase (LOX). When it reaches microenvironment, can be degraded release Mn2+. A Fenton-like reaction subsequently occurs generates large amount •OH around membrane. will increase permeability plasma for diffusion cells. The high concentration intracellular MDR. Owing coating, maintain excellent blood circulation accumulation abilities. In vitro vivo experiments have verified remarkable effect against tumors. This cell-mimicking strategy provides promising modality treatment.

Active Tumor Cell Resistance to Human Natural Killer Lymphocyte Attack1

Inhibition of protein synthesis by cycloheximide, puromycin, or emetine increased tumor lysis mediated by human natural killer (NK) cells to "slow" but not "fast" tumor targets. Human T24 bladder carcinoma cells were used as slow targets which are killed after a ~3-hr lag period, and K562 cells derived from a patient with myelogenous leukemia were used as fast targets which are killed without a...

Overcoming Multidrug Resistance in Human Cancer Cells by Natural Compounds

Multidrug resistance is a phenomenon whereby tumors become resistant to structurally unrelated anticancer drugs. P-glycoprotein belongs to the large ATP-binding cassette (ABC) transporter superfamily of membrane transport proteins. P-glycoprotein mediates resistance to various classes of anticancer drugs including vinblastine, daunorubicin, and paclitaxel, by actively extruding the drugs from t...

Overcoming multidrug resistance with nanomedicines.

INTRODUCTION Cancer remains the leading cause of death worldwide. Numerous therapeutic strategies that include smart biological treatments toward specific cellular pathways are being developed. Yet, inherent and acquired multidrug resistance (MDR) to chemotherapeutic drugs remains the major obstacle in effective cancer treatments. AREAS COVERED Herein, we focused on an implementation of nanos...

The Efficacy of Toll-Like Receptors in Awakening Dendritic Cell/Natural Killer Cell System for Eradication of Tumors

Natural killer (NK) cells are effector cells of the innate immune system that exert direct cytotoxic functions. Ubiquitously-expressed toll-like receptors (TLRs) have been recognized as one of the major components promoting dendritic cell (DC) maturation, which may induce polarized immune responses beneficial to cancer immunotherapy. TLR-activated NK cells and DCs are prerequisite for robust ac...
